These two functions both lead to endocytosis and may lead to degradation of the receptor which is the most important mechanism to attenuate EGFR signaling[6]. Upon EGFR stimulation Eps15 undergoes two post-translational modifications. The phosphorylation of Tyr850 [7] and the ubiquitination [8] of a yet unknown residues. The phosphorylation of Eps15 is required for internalization of EGFR[7] but not for Ubiquitination of Eps15[9]. Ubiquitination of Eps15 has a role in EGF receptor internalization but the exact role the ubiquitin moiety plays in this system is not yet known. The UIMs of Eps15 are required for Ubiquitination of the Eps15[9]. Recent studies claim that eps15 may bind to its own ubiquitin moiety with the UIM and subsequently disassociating Eps15 from EGFR and AP-2 clathrin complex.[10]
