Rafael Nadal will begin his quest for an Olympic gold medal against Italian Potito Starace, while top seed Roger Federer faces a tough opener against Russia's Dmitry Tursunov after the draw for the tennis event.
Wimbledon and French Open champion Nadal of Spain, who will overtake Federer as world number one on August 18, appears to have the tougher half of the 64-strong draw which also includes Serbian Novak Djokovic and in-form Briton Andy Murray.
Federer's opponent Tursunov, ranked 35, won his fourth career tournament in Sydney this year and was runner-up in Indianapolis last month.
But Federer, who has made the Olympics one of his top priorities this season, will be familiar with his powerful game after they played a practice match this week.
The Swiss star also holds a 2-0 career advantage over the Russian while Nadal has won all of his four meetings with Starace and and is yet to drop a set.
The British number one Murray, who won his first Masters title in Cincinnati last week to climb to a career-high ranking of six in the world, faces Taiwan's Lu Yen-Hsun in his opening match.
The 21-year-old Scot is scheduled to play Nadal in the quarter-finals, while Djokovic would be a likely semi-final opponent.
Third seed Djokovic plays Robby Ginepri of the United States.
Murray and his brother Jamie face Canada's Daniel Nestor and Frederick Niemeyer in the opening round of the doubles tournament.
Defending champion Nicolas Massu of Chile, currently ranked 125, received a kind draw against Belgium's Steve Darcis.
Massu and Fernando Gonzalez will open the defence of their doubles crown against Marin Cilic and Ivan Ljubicic, with a likely second-round clash against Federer and Stanislas Wawrinka.
Nadal and Tommy Robredo are on a quarter-finals collision course with top seeds Bob and Mike Bryan, followed by a possible semi against Federer and Wawrinka or experienced Indian duo Mahesh Bhupathi and Leander Paes.
The Olympic tennis tournament starts on Sunday and finishes a week later on August 17.

OLYMPIC absentee Andy Roddick confessed on Tuesday that he will surely have second thoughts about his decision to skip the Summer Games when the action begins this weekend in Beijing.
"I'm sure when I'm watching the (Friday) opening ceremony on television, I'll miss it," said the top seed at the ATP Los Angeles Classic.
"Of course a part of me will want to be there."
American Roddick decided weeks ago to skip the Olympics in favour of devoting his hardcourt summer to preparing for the August 25 start of the US Open, which follows immediately.
Fifth-seeded Russian Marat Safin, who advanced over American John Isner 6-3, 6-4 had opposite sentiments about missing the global sporting event.
"I don't care about the Olympics," said the 28-year-old who attended both Sydney and Athens, winning just one match on aggregate. "It doesn't make sense to fly 15 hours over and 15 hours back for one event."
Roddick's summer campaign has so far been a flop, with a second-round loss in Toronto and a no-show on court in Cincinnati, where he pilled out last week with a stiff neck.
"It was a rough decision not to go," he said of his Olympic choice. "But whichever way I chose I would be wrong. Still, my injury may be a blessing in disguise.
"I have two weeks (LA plus Washington) to get my game ready. If I'd played Beijing there would have only been one."
The American who said he absorbed his Olympic experience four years ago on Athens by attending other events and meeting fellow competitors, added that he is feeling good about his chance in LA despite only practising for two days.
